主要功能(Main function):硅基底芯片,其上嵌刻的水平通道可形成化学趋化因子浓度梯度,用于测定浓度梯度依赖细胞的功能,如趋化,脱颗粒。水平通道的深度小于悬浮细胞的直径,其内可观测细胞形态学变化和增殖迁移过程。细胞趋化分析不仅包括中性粒细胞、嗜酸性粒细胞、单核细胞、淋巴细胞等外周血白细胞,也包括各种癌细胞和培养细胞,如平滑肌细胞、内皮细胞、神经细胞、干细胞等。还可分析蛋白质及细胞相互作用、细胞信号转导、细胞骨架、钙流入、活性氧代谢等。可应用于趋化因子及药物筛选、炎症、过敏反应、肿瘤、神经、免疫、心血管、干细胞等方面的研究。(Silicon chip base, engraved on the horizontal channel can form chemical concentration gradient of the chemokines, for assay of concentration gradient-dependent cellular functions, such as chemotaxis and degranulation. The chip terrace depth is less than the diameter of the suspended cells, which morphology, proliferation and migration process can be observed. The cells for chemotactic analysis include not only leukocytes from peripheral blood such as neutrophils, eosinophils, monocytes and lymphocytes, but also cancer cells and cultured cells such as smooth muscle cells, endothelial cells, nerve cells and stem cells. Also for assays of proteins and cells interactionscell signal transductioncytoskeletal organization, calcium influx and active oxygen generation etc. Application to fields of chemokines and drug screening, inflammation, allergic reaction, oncology, neurology, immunology, cardiovasology and stem cell etc.
